Neuronal cell death in the mammalian retina occurs as a natural part of postnatal development, during the course of aging, and following axonal injury. The roles of cell death processes in the control of normal retinal development and in the maintenance of neural function during normal aging and following axonal injury have not been well understood. In this study, the effect of the bcl-2 gene, an inhibitor of apoptosis, on ganglion cell death was examined following axonal injury. Ganglion cells in whole-mount retinas of 3-day-old Sprague-Dawley rats were retrogradely labeled by injection of fast blue through a guide tube into the superior colliculus (SC). A single unilateral optic nerve transection was performed at 3, 7, or 14 days after the retrograde labeling. At 5 days after the transection, animals were treated with either adenoviral bcl-2 (Adbcl-2, 5 x 10(9) plaque-forming units/ml), or vehicle (control). At 15 days after the transection, animals were killed, and cell death in the SC was quantified. Retinal ganglion cell death increased significantly at 7 days after optic nerve transection, but no significant cell death was observed at 3 or 14 days after transection. Treatment with Adbcl-2 significantly inhibited ganglion cell death in the SC at all time points.
